SPE-HPLC Determination of Free Cortisol Levels in Human Serum

Abstract

A simple and sensitive analytical method is described for the determination of free cortisol in human serum using SPE-C18 clean-up and enrichment,coupled with high performance liquid chromatography with UV-detection.The mobile phase was methanol,aceticamine(0.02 mol/L),acetic acid and triethylamine(TEA),and volume ratio was 60∶40∶0.5∶0.5.The calibration curve of cortisol was linear(r=0.997 9) in the range of(0.1~3.0)×10-6 g/mL.The detection limit was 2.5×10-8 g/mL,and the recovery of cortisol was 86.0%~91.7%.The method was used for the analysis of cortisol in eight blood samples before and after narcotism.The proposed method is precise and suitable for the detection of free crotisol in serum.
