Objective Assay of the levels of Th1/Th2-types cytokines(IFN-γ/IL-4) in culture supernatants of peripheral blood mononuclear cells(PBMC) of patients with chronic hepatitis B,investigates the role of Th1/Th2-type cytokines in chronic hepatitis B.Methods The levels of IFN-γ and IL-4 in culture supernatants of PBMCs from 64 patients with chronic hepatitis B and 20 normal controls were assayed by ELISA method.PBMC were cultured with PHA(100μg/ml) and recombinant HBcAg/HBeAg(1.0μg/ml) for 48 hours in vitor respectively.Results ①The levels of IFN-γ produced by PHA-stimulated PBMCs from patients with chronic hepatitis B were much lower than that from control,but the levels of IL-4 was significantly higher than that in control.Moreover,the level of IFN-γ of PBMC in ALT abnormal patients with chronic hepatitis B was higher than ALT normal patients.②Compared with control group,IFN-γ expression was predominate when PBMC of patients with chronic hepatitis B were stimulated by HBcAg,in contrast,IL-4 expression was predominant when PBMCs were stimulated by HBeAg.Conclusion The date suggest that Th1-type cytokines production is associated with hepatic inflammatory activity of chronic hepatitis B and Th2-types cytokines production may be associated with chronics in HBV infection patient;HBcAg tend to induce Th1-type cytokines production and HBeAg tend to induce Th2-type cytokins pruduction in chronic hepatitis B.
