Abstract

AIM To study biological effects of antisense RNA on ovarian cancer cells. METHODS Ovarian cancer cell strain 8910 was transfected by CD147 antisense RNA expression vector PCDNAasCD147. Positive clones, selected and identified, were picked out for gelatin zymography, cell invasion assay and were transplanted subcutaneously in nude mice to observe the tumorigenesis. RESULTS Gelatin zymography showed that the capability of stimulating fibroblast to secret MMP-2 and MMp-9 was suppressed in 8910 transfected by PCDNAasCD147 compared with that of the non-antisense group. Invasion and tumorigenesis in nude mice of 8910transfected by PCDNAasCD147 were lowered. CONCLUSION CD147 antisense RNA can inhibit invasion and tumorigenesis of ovarian cancer cells and CD147 may become a new drug target of tumor therapy.
