Abstract

Objective To discuss the feasibility of establishing a model of chronic bronchitis in rats by inhaling cigarette.By observing the changes of ultrastructures of bronchus,the effects of ambroxol on rats with chronic bronchitis were observed.Methods Rats were randomly divided into three study groups:normal control group,smoking model group and ambroxol intervention group.The rats model of chronic bronchitis were established by inhaling cigarette.For the ambroxol intervention group,ambroxol was given 40 mg·kg-1·d-1 for 2 months before the rats were put to death.The characteristics of ultrastructures of airway inflammation were measured,and the bronchoalveolar lavage fluid (BALF) cell count and differential count were measured.Results The ultrastructure changes of the model were consistent with that of patients with chronic bronchitis.The pathological changes of the air tube and the ultrastructure of bronchial epithelial cells in the ambroxol intervention group remained compared with normal control group,but improved obviously with smoking group.In BALF,the total cellular score and mononuclear macrophage percentage in the smoking group and intervention group were significantly different compared with normal control group(P0.05).There was significant difference in total cellular number between smoking group and intervention group(P0.05).Conclusions Establishing the chronic bronchitis model in rats induced by inhaling cigarette is feasible.Long-term application of ambroxol can protect the airway of rats with chronic bronchitis.
