Abstract
Objective:To investigate the effect of mycophenolate mofetil(MMF)on high glucose-induced proliferation and the type Ⅳ collagen in rat mesangial cells(MCs).Methods:Rat MCs were stimulated by different concentrations of high glucose,out of which 30?mmol/L was fit for the stimulation concentration,and then different concentrations of MMF were added into MCs of glucose induced rat.The MCs proliferative rate within 6,12,24 and 48 hours was detected by MTT colorimetry.The type Ⅳ collagen in supernatant contents of medium was detected by radioimmunoassay method.Results: The MCs proliferation presented time-and dosedependence by different concentrations of high glucose,which got to the best at the concentration of 30?mmol/L and at 24 hours.MMF could inhibit the high glucose-induced proliferation and showed time-and dose-dependence.Conclusion: High concentration of glucose can stimulate MCs and results in MCs proliferation by time-and dose-dependence,meanwhile MMF can inhibit such a high glucose-induced proliferation of MCs,also showing time-and dose-dependence.
