Abstract
Aim: To observe the effects of immune treatment of dendritic cells on S180 bearing mice with splenectomy and study the relationship between spleen and DC in antitumor immune response. Method:A total of 33 healthy Kunming mice were randomly allocated into 3 groups:operation control group, splenectomy group and splenectomy+DC group. In the 8th week S180 were injected hypodemically in the sides of crural oxter respectively in 5 mice of each group, 2×10 6 cells per mouse, or injected into abdominal cavity in 6 mice of each group, 1×10 6 cells per mouse. S180 lysate pulsed dendritic cells were injected into abdominal cavity,4×10 4 cells per mouse,and repeated after 1 week . The tumor size under skin and the body weight were measured. Results:The immune effects of DC in peritoneal S180 bearing mice with splenectomy showed that the growth velocity of ascite and death time between 3 groups had no significant differences ( P 0.05);the immune effects of DC subdemal S180 bearing mice with splenectomy showed that the tumor incidence rate of the splenectomy group had no significant difference compared with that of operation control group( P 0.05), but the death time of most mice (4/5) were less than that of the operation control group. The survival time of splenectomy mice with DC treatment had significantly prolonged, survival time of 80%(4/5) mice exceeded 60 d,while the operation control group and splenectomy group were 60% (3/5) and 40% (2/5),respectively. Conclusion:The survival time of splenectomy mice decreases; DC may induce antitumor immune reaction for splenectomy mice bearing S180 in certain time.
