Growth inhibition effect of small interfering RNA on tumor growth of human gastric caner cell SGC7901 targeting Akt1

Abstract

Objective To investigate growth inhibition effect on tumor growth of gastric cancer cell SGC7901 by small interfering RNA targeting Akt1.Methods SiRNA targeted human Akt1 was constructed,gastric cancer cell SGC7901 cultured and transfected with Akt1 siRNA Western blot and real time PCR were used to examine the silence result.Flow cytometry and Transwell assay were used to detect proliferation,invasion and apoptosis.Results The results showed that the expression of Akt1 was down-regulated in Akt1-siRNA group.And G0/G1 arrest was observed in Akt1-siRNA group(χ2=11.076,P=0.026);Apoptosis cells increased when Akt1 was down regulated(F=89.386,P=0.00),and cell invasion activity was inhibited dramatically(F=21.351,P=0.00).Conclusions SiRNA targeted Akt1 specially down regulate expression of Akt1,inhibited gastric cancer cell growth and invasion activity,induced cell apoptosis.SiRNA approach should be a new strategy in gastric cancer gene therapy targeting Akt1.
