Effect of D-jglucose on the Extracellular Matrix of Mesangial Cells

Abstract

Objective To study the effect of different levels of high glucose on extracellular matrix mesangial cells in different time.Methods Mesangial cells were cultured in different levels of D-glucose (5.5,10,20,30 mmol/L)in different time(6,12,24,48 h).The levels of fibronectin and collagen Ⅳ were determined by enzyme linked immunosorbent assay (ELISA) in the culture media.Results (1) Among the levels of 10,20,30 mmol/L D-glucose,the levels of fibronectin and collagen Ⅳ were increased according to the extension of cultured time (6,12,24,48 h) (P0.05),the level of fibronectin and collagen Ⅳ of 24 h were higher than those of other time (6,12,48 h) (P0.05).(2) On the same time,the level of fibronectin and collagen Ⅳ were also raised according to the increase of the levels of D-glucose(5.5,10,20,30 mmol/L)(P0.05),the levels of fibronectin and collagen Ⅳ of 30 mmol/L D-glucose were higher than those of other levels of D-glucose(5.5,10,20 mmol/L)(P0.05).Conclusion D-glucose can promote mesangial cells to produce extracellular matrix according to time and dose mode.
