Abstract
Aim:To investigate the injuries of hepatocyte and sinusodial endothelial cell(SEC)during different hepatic cold preservation and reperfusion period.Methods:Sixty-three Wistar rats were randomly allocated into 5 groups:4 experimental groups(E1,E2,E3,E4),14 rats in each group,7 as donors and 7 as recipients,and the 7 else was the normal control(group N).Liver grafts in groups E1,E2,E3,and E4 group were flushed with and preserved in 4 ℃ Euro-collins solution,the cold preservation time was 3 h,6 h,8 h,and 12 h,respectively.Then orthotopic liver transplantation was performed.At 15 min,30 min,and 60 min after portal vein reperfusion,blood samples were obtained to determine ALT,AST,ET and HA.60 min after portal vein reperfusion,the ultrastructure of hepatocyte and SEC of grafts samples were observed.Results:The serum levels of HA and ET at 15 min after portal vein reperfusion in E1,E2,E3,E4 groups began to increase compared with those in group N(P0.05),and the levels of HA and ET increased gradually along with the time of reperfusion and prolongation of cold preservation.At 30 min after portal vein reperfusion,the level of ALT and AST in E1,E2,E3,E4 groups began to increase compared with those in group N(P0.05).Hepatocyte of grafted liver in group E1 and E2 were normal,but that in group E3 and E4 were injuried.SEC injury appeared in all experimental groups,and the injury degree of SEC were more severe than hepatocyte.Conclusion:During cold preservation and reperfusion,the appearance of SEC injury was earlier and more severe than hepatocyte.
