Abstract
Objective:To explore the effect of Benazepril on cardiac ACE2 gene and protein expression in rat congestive heart failure de- rived from myocardial infarction. Methods:The rat heart failure model was induced by acute myocardial infarction(AMI)by ligating the left anterior descend- ing coronary artery.One month after operation,rats were randomized to CHF placebo group and Benazapril group.Sham-operated rats were used as the control group.The following indexes were evaluated at 8th week:①Hemodynamics;②Body mass and left ventricular mass index,③The circulating and cardiac level of angiotensinⅡ,④ACE2 gene and protein expression using RT-PCR and Western Blot. Results:The cirlutating and cardiac angiotentinⅡlevel were increased significantly in CHF placebo group than those in sham-operation group(P0.01).The ACE2 mRNA and protein levels were increased in CHF placebo group than those in sham- operation group,respectively(P0.05,P0.01).The bemodynamic parameters in Benazapril group improved significantly than those in CHF placebo group(P0.01).The LVMI in Benazapril group was decreased obviously compared with that in CHF pla- cebo group(P0.01).The level of ACE2 mRNA and protein were increased significantly in Benazapril group(P0.01). Conclusions:ACE2 gene and protein expression were upregnlated in CHF rats.Benazapril can further upregnlate the expres- sion of ACE2.This might be the new explanation of the mechanism of the drug.
