Abstract
Objective:To investigate the internalization how to regulate the signal transduction of HGF-cMet,specially the influence of Dynamin on internalization of HGF-cMet by receptor inducing.Methods:Hepatocyte growth factor was labeled with ~(125)I.The quantity of HGF internalization of ~(125)I HGF in HepG2 cell at 4℃ and 37℃ and different time point was compared(respectively) with cell uptake assay.Results:Internalization of ~(125)I-HGF in HepG2 cell was reduced about 63% in cell line of K44A mutation Dynamin transfected at 37℃.At 4℃,the level of interlization maintained very low.Conclusion:The process of signal transduction was induced by HGF receptor cMET and was related with internalization of cell.The danamin is important role in process of internalization.
