Protective Effect of Cardioplegia with Diazoxide to Myocardial Mitochondrion during Ischemic and Reperfusion in Isolated Working Rat Heart

Abstract

OBJECTIVE To evaluate the effects of mitochondrial ATP-sensitive potassium channel opener diazoxide on heart mitochondrion during ischemic and reperfusion in rats.METHODS Fifty rats were divided into 5 groups randomly.Langendorff model was established.Control group(B) underwent 30 minute ischemia and 60minute reperfusion.Group C and D were treated with St.Thomas Ⅱ cardioplegia and diazoxide+St.Thomas Ⅱ cardioplegia before ischemia respectively.And group E was treated with glibenclamide at 10min before Diazoxide+st.Thomas Ⅱ cardioplegia.At the end,mitochondrial changes were detected under the transmission electron microscope(TEM),the respiration function of mitochondrion was determined,ATP,ADP,AMP content of myocardium were determined and energy charge(EC) were calculated.RESULTS Ischemic and reperfusion injury induced the structure damaged and function decrease of myocardial mitochondrial.While in group D(P0.01),the heart was protected effectively by treating with diazoxide.CONCLUSION These results suggest that diazoxide has a protective effect on myocardial mitochondrion during ischemic and reperfusion in rats.
