Abstract

Objective: To investigate the inhibitory effect of antisense oligodeoxynucleotide(AODN)as a specific inhibitor of CVB 3 replication on CVB3 pathogenicity in vero cells and in Balb/C mice. Methods: Cytopathic effect(CPE)was investigated,50% tissue culture infection dose(TCID50)was titrated on vero cells monolayer and cardiac histological lesions were investigated. Results:The specific AODN could significantly inhibit CPE of CVB3_infected vero cells and decrease viral titres in culture supernatant in dose dependent manner.The specific AODN could also significantly decrease viral titres of myocardial tissue,lighten cardiac lesions and reduce incidence rate of myocarditis in mice in dose dependent manner. Conclusion: AODN may be protect vero cells and myocardial tissue against CVB3 infection by inhibiting CVB3 replication.
