Isolation and Purification of Two Saponins from Semen celosiae by High-speed Counter-current Chromatography Coupled with ELSD

Abstract

High-speed counter-current chromatography(HSCCC) coupled with evaporative light scattering detection(ELSD) was successfully applied to preparative separation and purification of two saponins,celosins A and B,which were from the main saponins of Semen celosiae by a one-step separation,using dichloromethane-methanol-n-butanol-water(4∶ 3∶ 0.3∶ 2,v/v/v/v) with addition of 0.5% glacial acetic acid as elution solvent..HPLC analysis of the fractions collected from the semi-preparative HSCCC of 100 mg of the main saponins showed that the purity of celosin A(30.5 mg) was 98.9% and that of celosin B(10.7 mg) was 98.1%.The chemical structures of these components were confirmed by 13C NMR and EI-MS.
