Abstract
Pectinase was immobilized on anion-exchange resin by cross-linkage of glutaraldehyde to study the effects of temperature,pH,time,amount of the enzyme,concentration of glutaraldehyde,crosslinking temperature and time on pectinase's immobilization and the characteristics of immobilized enzyme.The optimum conditions for the immobilization were as follows:temperature 40°C,pH 5.5,reaction time 6 h,quantity of pectinase 0.75 mL/g,concentration of glutaraldehyde 0.1%,crosslinking temperature 4°C and crosslinking time 4 h respectively.Moreover,the immobilized pectinase showed a good operational stability.
