Culture and Differentiation of Neural Stem Cells from Spinal Cord of GFP-Transgene Embryonic Rat and Expression of Neuregulin-1

Abstract

Aim:To isolate and culture the neural stem cells(NSCs) from spinal cord of the GFP+-transgene embryonic rat,and to observe the multiple functions and Neuregulin-1 expression of NSCs. Methods:NSCs were isolated and cultured from spinal cord of the GFP+-embryonic rat(E16d) . The Neuregulin-1 expression of neurospheres and identification of cellular types differentiated from NSCs were determined by the method of immuno-histochemistry. Results:NSCs could be isolated and cultured from spinal cord of the GFP+-embryonic rat. The neurospheres could express Neuregulin-1 and Nestin,and it could further differentiate into neurons,astrocytes and olygodenrocytes. Conclusion:NSCs could be isolated and cultured from spinal cord of the GFP+-embryonic rat. The NSCs have multiple functions of which differentiate into various neurocytes used in the treatment of CNS diseases.
