Abstract
Objective To observe the effects of ketamine on the expression of heme-oxygenase-1 in lung tissue of sepsis in rats and explore the possible action mechanism.Methods 40 male vista rats were randomly divided into 5 group: sham group(group A),CLP group(group B),ketamine 40mg/kg group plus CLP(group C),ketamine 40mg/kg plus zinc protoporphyrin Ⅸ 5mg/kg plus CLP(group D),zinc protoporphyrin Ⅸ 5mg/kg plus CLP(group E).Rats were killed at 9h after CLP.The levels of MDA in lung tissue were measured,bronchoalveolar lavage fluid(BALF) was collected for measurement of concentration of total protein.HE staining was used to observe pathological changes of lung.And the level of HO-1 was measured by immunohistochemical method.Results Compared with group A,the contents of MDA,TP and the expression of HO-1 were increased in other injury groups(P0.01);compared with group B,the contents of MDA,TP were decreased and the expression of HO-1 was up-regulated in group C(P0.05 or P0.01);compared with group C,the contents of MDA,TP were increased and the expression of HO-1 was down-regulated in group D(P0.01);compared with group B,there is no significant differences in the contents of MDA,TP,and the expression of HO-1 in group D,E.Conclusion Ketamine extenuate lung injury in sepsis rats through up-regulating HO-1 expression.
