High-level expression of anti-CEA chimeric antibody in CHO-dhfr cells

Abstract

Objective To express chimeric anti-CEA ant ibody in dihydrofolate reductase-deficie nt Chinese hamster ovary(CHO-dhfr) cells at high-level.MethodsThe euka ryotic expression vectors of chimeric anti-CEA antibody were constructed and ind ucted into CHO-dhfr- cells.High-level expression was achieved after cells adapted to methotrexate.Chimeric antibody was characterized by ELISA,Western blot,and radioimmunoassays in vivo.Results The cell strain that secrets anti-CEA chime ric antibody at high-level of 60 μg/ml w a s established.It was found that the chimeric antibody contained constant regions of human Ig and had the specificity against CEA by ELISA,Western blot,and radi oimmunoassays in vivo.Conclusi on The anti-CEA mouse-human chimeric antibody was successfully expressed at high-level in CHO cells.
