Abstract
Objective To research the effect of selenium on expression of inducible nitric oxide synthase(iNOS) and nitric oxide synthase(eNOS) induced by arsenic in HUECV-304(human umbilical vein endothelial cells).Methods The density of HUECV-304 was adjusted to 1.0×105 /ml,and the cells were added respectively to final concentrations of 0.05,0.1,0.5,1.5,3.0,6.0 and 12.0 μmol/L arsenic,and 0.1 μmol/L selenium+0.05,0.1,0.5,1.5,3.0,6.0,12.0 μmol/L arsenic,exposed for 48 h,and the control(normal culture medium) group was established.The activities of iNOS and eNOS were determined,and the expression of iNOS mRNA and eNOS mRNA was analyzed by the RT-PCR.Results With the increasing of arsenic exposure,HUECV-304 showed a gradual decline trend in the activity of eNOS,and the activity of iNOS showed a gradual upward trend.Compared with control group,iNOS mRNA expression were higher,and eNOS mRNA expression were significantly lower in the 0.5-2.0 μmol/L arsenic exposure group(P0.05,P0.01).Compared with the corresponding concentration of arsenic exposure group,the activities of iNOS and iNOS mRNA expression were lower in the 0.05,0.1,1.5 μmol/L arsenic+0.1 μmol/L selenium combined exposure group,and the activities of iNOS and eNOS mRNA expression were higher in the 0.05,0.5,1.5 μmol/L arsenic+0.1 μmol/L selenium combined exposure groups(P0.05).Conclusion Selenium may inhibit the abnormal expression of iNOS and eNOS induced by arsenic in the vein endothelial cells when the level of the arsenic exposure is not too high.
