Effects of polydatin on platelet aggregation and platelet cytosolic calcium

Abstract

To investigate effects of polydatin on platelet aggregation and platelet cytosolic calcium, platelet aggregation was tested according to Borns method; platelet cytosolic calcium was determined by use of Fura-2/AM.In vitro polydatin significantly inhibited platelet aggregation induced by AA and ADP,and IC 50 values(concentration resulting in 50% inhibition) were 5.13 for AA and 10.07 μmol/L for ADP respectively.In vivo polydatin(5,10,20 mg/kg) could markedly inhibit rabbit platelet aggregation induced by AA and ADP.Polydatin markedly lowered the concentration increase in platelet calcium induced by thrombin through inhibition of both the influx of extracellular calcium and the mobilization of calcium from intracellular stores. These results showed that polydatin evidently inhibited platelet aggregation and decreased concentration of platelet cytosolic calcium.It is suggested that the antithrombotic effect of polydatin may be related to its anti-platelet aggregation and decrease of platelet cytosolic calcium.
