Abstract
Objective:To study the expressions of surface markers(CD83,CD80,CD86)in mononuclear derived dendritic cells orientational cultured in vitro in peripheral blood of patients with type Ⅰ diabetes and secretion of cytokines and changes of NF-kB activity in dendritic cells,explore the effect of NF-kB in pathogenesis of type Ⅰ diabetes.Methods:20 patients with diabetic were divided into IDM Group(10 cases)and oligodeoxyribonucleotide(ODN)group(10 cases)randomly,10 healthy volunteers without diabetic were selected as control group.Peripheral blood mononuclear cells(PBMC)were isolated in the three groups,then were induced and cultivated with IL-4 and GM-CSF,NF-kB specific oligodeoxyribonucleotide was added into ODN group to culture cells,the levels of surface markers(CD83,CD80,CD86)were detected by flow cytometry,the level of IL-12 in supernatant liquid was detected by ELISA.Results:The levels of IL-12 in supernatant liquid of control group and ODN group were significantly lower than that of IDM group(P0.05);there was no significant difference between control group and ODN group(P0.05).The expression levels of CD83,CD80 and CD86 in control group and ODN group were significantly lower than that of IDM group(P0.05);there was no significant difference between control group and ODN group(P0.05).Conclusion:Maturation and abnormal function of dendritic cells in patients with type Ⅰ diabetes are related to enhanced NF-kB activity.ODN may suppress NF-kB activity in dendritic cells and reduce the expression of costimulatory molecules and IL-12 secretion.
