Abstract

Objective To establish a method for the determination of alkaloids in extracts from Coptis by HPLC. Methods Epiberberine, coptisine, palmatine and berberine in extracts from Coptis were determined by HPLC. The column was Kromasil 100-5 C18, with the mobile phase consisted of acetonitrile-phosphate buffer(53:47, v/v) at the f low rate of 1.0 mL/min. The UV detection wavelength was 345 nm and the injection volume was 10 μL. Results The linear range of berberine hydrochloride was 0.1032-2.5800 μg(r =0.9999), with an average recovery of 100.19 %(RSD=1.77 %). Contents of alkaloids of six batches of extracts from Coptis were determined. The content of epiberberine, coptisine, palmatine and berberine was not less than 5.5 %, 11.5 %, 9.0 %, 44.0 %, respectively, and the content of total alkaloid was not less than 70.0 %. Conclusion This method is simple, reliable and accurate, and can be used for quality control of extracts from Coptis.
