Effects of Toll-like Receptor-2 on Interleukin-8 Secretion of Keratinocytes

Abstract

Objective To observe the change of Interleukin-8(IL-8) in keratinocytes induced with Candida Albicans and the effect of Toll-like receptor-2(TLR-2) on IL-8 secretion,to explore the role of keratinocytes in skin immune system.Methods Candida Albicans were cultured on keratinocytes treated before and after anti-TLR-2 monoantibody(mAb),respectively.The levels of IL-8 in culture supernatant were determined with ELISA at different time points.Results The IL-8 levels were significantly increased after induced with Candida Albicans(P0.01),the significant increase was observed at 3 h and reached the peak at 24 h;the increase of IL-8 levels had no significant difference after TLR-2 was neutralized with anti-TLR-2 mAb(P0.05).Conclusion The secretion of IL-8 in keratinocytes could be induced with Candida albicans,the function of TLR-2 is uncertain in expression of IL-8 in keratinocytes.
