Abstract
Objective To establish rat model with liver fibrosis induced by CCI4,and to evaluatethe role of interferonα-2a(IFNα-2a) in experimental liver fibrosis as well as its effect on the expression of TGF-β1,so as to explore its underlying mechanisms. Methods Forty-nine maleWistar rats were randomly divided into 5 groups.Except the normal control group, the 40% CCl4 olive oil solution was injected into abdominal cavity with 20% ethyl alcohols to fill the stomach to make the mold. After making the mold, the treatment group was treated with dif-ferent intramuscular injections of IFNα-2a fluid, while the control group was filled with the col-chicine.After 12 weeks, blood was drawn from femoral artery and PC Ⅲ level was examined to prepare the pathology specimen of hepatic tissue. Results As compared with the model group, the rats treated by high-dose IFNα-2a and colchicine gained weight, the serum levels of PCⅢ were significantly decreased(P 0.01),the liver fibrosis was significantly improved;the ex-pressions of TGF-β1 were decreased in the liver of the rats which were given high-doseIFNα-2a(P0.01).Conclusion It was proved that IFN α-2a could be effective in treatingCCl4 induced liver fibrosis in rats by affecting the expression of TGF-β1.
