Composition and Optimization of Extraction of Collagen from Navodon septentionalis Skin Using Response Surface Methodology

Abstract

The purpose of this study was to use response surface method to optimize the extraction of collagen from the Navodon septentionalis skin and analyze the composition of amino acid of the collagen. The study would provide some theoretical basis for the further processing and high-value utilization of Navodon septentrionalis skin. The results showed that the optimum pre-treatment conditions of collagen extraction were as follows: the condition of degreasing was 10% isopropyl alcohol,solid-liquid ratio 1 ∶ 15,treatment time 20 h; the condition of removing non-collagen proteins was 7. 5% NaCl,solid-liquid ratio 1∶ 10,treatment time 24 h. The optimum extraction condition was as follows: 0. 38 mol ·L- 1acetic acid,solid-liquid ratio 1: 30,extraction time 33 h. Under this condition,the yield of collagen protein was 362. 5mg·g- 1. The compositional features of the amino acid were with the high contents of Gly( 24. 12%) and Hyp( 9. 31%),but Try was undetected. It was concluded that the extracts were collagen.
