Effect of Yi-Shen-Tiao-Du Acupuncture-Moxibustion on Hippocampus Mitochondria of COXIV in Alzheimer's Disease Rat

Abstract

Objective: To observe the effects of Yi-shen-tiao-du acupuncture-moxibustion on the AD( Alzheimer's disease)rats with hippocampus mitochondria COXⅣ,and to investigate the specific mechanism of preventive-treatment on AD by acupuncturemoxibustion. Methods: Wistar rats were randomly divided into 4 groups: the normal group,sham-operated group,model group,and acupuncture-moxibustion group. First,bilateral hippocampus were injected Aβ1-42to make rat model of AD. Then,acupuncture-moxibustion group selected Baihui points and Shenshu points,and treated with acupuncture and moxibustion. Last,detect the expression of hippocampus mitochondria COXⅣ in rats by the method of immunohistochemistry. Results: With rat behavior-observed,the acupuncture-moxibusition group which reacted by external stimuli could still,and eating-drinking normally,compared with the model group improved. Meanwhile,through immunohistochemistry analysis of average pptical and average gray about the expression of COXⅣ in hippocampalnerons mitochondria,it could be found that compared with normal group and sham-operated group,COXⅣ expression of model group's hippocampus mitochondria is lower obviously than that of them( P 0. 01). However,compared with the model group,COXⅣ expression of acupuncture-moxibustion group's hippocampus mitochondria increased significantly( P 0. 01). Conclusion: Yi-shen-tiao-du anupuncture-moxibustion can increase the activity of hippocampapal mitochondrial respiratory chain enzyme complexe and ATP concentriation improve mitochondrial function. Thereby Yi-shen-tiao-du acupuncture-moxibustion plays a role in treatment and prevention for Alzheimer's disease.
