Detection of VEGF mRNA in Breast Carcinoma by Real-time Fluorescence Quantitative RT-PCR

Abstract

Objective:To establish a real-time fluorescence quantitative RT-PCR for detection of VEGF mRNA expression in breast carcinoma,and verify the correlation with protein expression detected by the immunohisochemistry LSAB method.Methods:Total RNA was extracted with Trizol from the tissues and mRNA was transcribed reversely into cDNA.The real-time quantitative RT-PCR was used to detect the expression of VEGF mRNA in 44 invasive ductal breast cancer,25 control breast tissues and 13 benign breast diseases.The immunohistochemistry LSAB method was used to evaluate the protein expression of VEGF.Results:The expression of VEGF mRNA was significantly higher in the breast carcinoma tissues than that in the normal breast tissue and benign tissues(P0.001),while no significant difference was found in the latter two tissues(P0.05).The expression of VEGF mRNA was correlated with that of the VEGF protein significantly.Conclusion:Real-time fluorescence quantitative RT-PCR used to detect the expression of VEGF mRNA might be a reliable method for early diagnosis of breast carcinoma.
