Abstract
To investigate the effects of apoptosis of bone marrow stromal cell line HS- 5 on acute myeloid leukemia cell line HL- 60.The experiments were divided into two groups:direct contact co- culture of HS- 5 cells and HL- 60 cells(HS- 5:HL- 60=1×105/m L:1×106/m L)(experiment group) and HL- 60 cells(1×106/m L) cultured alone(control group).Flow cytometry(FCM) was used to evaluate the early apoptosis rates of HL- 60 cells in 24、48、72 h;Observing the changes of morphology of HL- 60 cells through Wright's stain;reverse transcription PCR( RT- PCR) was used to detect the expression of anti- apoptosis genes bcl- 2 and bcl- xl.As the results showed that FCM showed that the apoptosis rates of HL- 60 cells of experiment and control groups have no statistically significance differences at 24 h and 72 h(P0.05).While the apoptosis rates of HL- 60 cells remarkably reduced compared with control group at 48 h(P0.05).The morphology of HL- 60 cells of control group was more irregularthan that in the experienment group through Wright's stain.At the same time,the anti- apoptosis genes bcl- 2 and bcl- xl were up- regulated(P0.05).From above,it is concluded that direct contact co- culture of HS- 5 cells and HL- 60 cells which can protect HL- 60 cells from apoptosis rapidly,which is partly due to the anti- apoptosis genes bcl- 2 and bcl- xl are up- regulated.
