Abstract
Objective To study the mechanisms of TTF1 inhibiting the proliferation of hepatocellular carcinoma(HCC) cells. Methods The inhibitory rate of TTF1 on HCC cells was detected by MTT on the normal human liver cell lines,Chang-liver,and the HCC cell line,HepG2.The effect of TTF1 on HCC cells proliferation was detected by EdU assay.And the protein expression levels of wild type p53 and Bcl-2 were examined by western blot. Results The MTT and EdU data showed that TTF1 inhibited HCC cell line HepG2 proliferation.Western blot analysis showed that TTF1 treatment led to decreased protein expression of bcl-2 and an increase in wild type p53. Conclusion These results together suggested that TTF1 may induce proliferation of HepG-2 cells through a complex pathway.
