The observation of skin explants culture to harvest primary cell

Objective To harvest primary cells from skin explants culture. Methods Skin explants in size of 2mm×2mm were obtained by using sharp scissors,each skin explant placed onto the bottom of culture dish,and they were oriented with the epidermis facing up,then observe the cells migrating from the explants. Results From the third day,keratinocytes migrating from the explants,then melanocytes appear around the explants.As culture time getting longger,a large number of fibrolasts around the explant. Conclusion During the explant culture,we could harvest keratinocyte without contamination firstly,and DMEM contain serum culture for 20 days,could harvest a large number of fibrolasts.
