Abstract
Objective To explore the inhibitory effect of resveratrol(Res) on transplanted cervix cancer cell U14 in mice,and its related molecular mechanisms.Methods The 615-strain mice with U14 cervical cancer cells were divided into three groups randomly,namely control group(n=10),Res group(n=10) and cisplatin group(n=10).After drug intervention,all mice were sacrificed on day 26 posttransplantation.The volumes and weighs of tumors were detected,and tumor inhibition rate was calculated.Microvessel density(MVD) was measured by immunohistochemistry.The levels of hypoxia inducible factor-1α(HIF-1α) as well as vascular endothelia growth factor(VEGF) mRNA and protein expression were measured by real-time quantitative PCR and Western blotting,respectively.Results The tumor inhibition rate was 50.46% and 57.19% in Res group and cisplatin group,respectively.The volumes and weights of tumor was markedly decreased in Res group and cisplatin group compared with control group(P0.01).The levels of HIF-1α as well as VEGF mRNA and protein expression and MVD was significantly lower in Res group than that in control group respec tively(P0.01).However,there was no significant difference in above indicators between Res group and cisplatin group(P0.05).Conclusion Res may suppress the growth of cervix cancer by downregulating the expression of HIF-1 α and VEGF in mice.
