Isolation and Identification of Four Highly Effective Phenol-degrading Strains from Coking Wastewater

Abstract

Objective:To isolate and identify highly efficient phenol-degrading strain from coking wastewater.Method:The microbes were isolated by the inorganic salt medium containing 100-1 000mg/L phenol as a sole carbon source in order to obtain efficient phenol-degrading strain.The growth curve and phenol-degrading efficiency was determined,also the taxonomic statuses of these strains were determined based on morphological features and 16S rDNA sequences analysis.Result:Four highly effective phenol-degrading bacterial strain named ZL1,ZL2,ZL3,ZL4 were isolated and identified,while strain ZL1 belongs to Bacillus cereus,ZL2 and ZL4 belongs to Alcaligenes faecalis,ZL3 belongs to Serratia sp.The four bacterial strain could utilize phenol as a sole carbon,tolerate phenol of 1 000mg/L,and degrade phenol of 1 000mg/L above 91% in 24h when growing on the inorganic salt medium at temperature 30℃,pH 7.0 and 2% inoculation amount.Conclusion:Four highly effective phenol-degrading bacterial strain were obtained and have potential application in the biodegradation of high concentration organic wastewater with phenol substances.
