Effect of goat testis extract on the expression of Bcl-2 and Bax in mouse spermatogenic cells with lead exposure

Abstract

Objective To investigate the effect of goat testis extract on the expression of apoptosis-related gene Bcl-2 and Bax in mouse spermatogenic cells with lead exposure.Methods Thirty adult male Kunming mice were divided into three groups:normal saline;experimental group one,lead acetate;experimental group two,lead acetate+goat testis extract,which was given saline,lead acetate and lead acetate+goat testis extract.Respectively,once a day,for 14 days.7days after stopping injection,the mice were anaesthetized and testis were taken out.Protein expression level of Bcl-2,Bax in testis was detected by immunohistochemieal assay accordingly.Results Compared with the control group,protein expression level of Bcl-2 in experimental group one was lower(P0.01),and protein expression level of Bax was significantly higher(P0.01).No significant difference between experimental group two and control group(P0.05) was found.Conclusion Lead could accelerate the apoptosis of spermatogenic cells.Administration of goat testis extract could inhibit spermatogenic cell apoptosis by increasing Bcl-2 expression and decreasing Bax expression.
