【Objective】 To identify the differentially expressed proteins in T24 and BIU-87 human bladder transitional carcinoma with different malignancy by comparative proteomics techniques.【Methods】 Total proteins were extracted from human bladder cancer cell lines BIU-87 and T24 cultured in vitro.The proteins were then resolved by two-dimensional gel electrophoresis(2-DE) and stained with coomassie brilliant blue to find the differentially expressed proteins,which were analyzed by matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF-MS) and bioinformation.【Results】 153 protein spots were found over 2-fold change in expression by comparing the protein expression profiles of T24 and BIU-87,94 of which were analyzed by mass spectrometry and peptide mass fingerprinting(PMF).The protein expression of Rho GDP-dissociation inhibitor 1,Cathepsin D,CK-9,ACTG1 protein,MN-SOD,heat shock protein,ACTB and Thioredoxin domain-containing protein 4 were found up-regulated in T24;the expression of 14-3-3δprotein,orphan hormone nuclear receptor ROR alpha 3 was down-regulated in BIU-87.【Conclusions】 The changes in protein expression of 153 proteins occurred in T24 and BIU-87 human bladder transitional carcinoma.Further analysis of these differentially expressed proteins can be potential tumor-related candidate biomarkers.
