Abstract
Objective To study the modulating action on the fibrinolytic system,of thrombospondin 1 (TSP 1) and urokinase typed plasminogen activator (uPA) that express in remnant renal tissues of 7/8 nephrectomized rats during their tubulointerstitial fibrotic lesions.Methods 7/8 nephrectomized SD rats served as experimental animal model,After treated for 12 weeks with combination of Hirudo and Lotensin,rats were sacrificed and renal morphological changes were detected by use of PAS staining.The protein expression of TSP 1 and uPA in areas of remnant renal tubulointerstitium,the correlation between both proteins and TSP 1 with tubulointerstitial fibrotic lesions were estimated by means of immunohistochemical technique.Results The progressive glomerulosclerosis,tubulointerstitial fibrosis and chronic renal failure developed gradually after 7/8 nephrectomy in rats.But following the treatment,the renal functional parameters and pathological lesions ameliorated more significantly than untreated group.The immunostaining suggested that the expression of TSP 1 and uPA in the regions of tubulointerstitium was up regulated,and these two proteins′expression had showed a positive correlation on a time location dependent pattern,but a negative correlation had presented between TSP 1 and tubulointerstitial lesions.Conclusion The expression of TSP 1 and uPA in remnant renal tissues may be play an importment role in enhancing the fibrinolytic function during the restoration stage of tubulointerstitial lesions after treatment.
