The study of preventing autologous vein graft restenosis by smearing ligustrazine on the testing vessel adventitia

Abstract

Objective To investigate the effect of smearing ligustrazine locally on the preventing of vein graft failure and its mechanism.Methods Rabbit models of jugular veins grafted into the common carotid arteries were prepared.Animals were randomly divided into pluronic-F-127 gel group(smearing pluronic-F-127 gel on the adventitia of autologous vein grafts) and ligustrazine group(smearing ligustrazine mixed with pluronic-F-127 gel on the adventitia of autologous vein grafts).BrDU were localized by immunohistochemical procedure 3 days,7 days and 14 days after the operation.Fourteen days after the operation,the thickness of neointima,media and neointima/media ratios(I/M) were calculated from the sections stained with HE and Masson by computer image analysis system.Results Fourteen days after the operation,the thickness of neointima,media and I/M ratios in ligustrazine group had significantly changed comparing to pluronic-F-127 gel group:(1)the thickness of neointima was decreased by 68.35%.(2) the thickness of media was decreased by 22.02%.(3) I/M ratios was decreased by 54.48%.From 3 days to 14 days compared to pluronic-F-127 gel group,the proliferating rate of vessel in ligustrazine group decreased by 31.78%,41.03%,38.75%,respectively.Conclusions Smearing ligustrazine mixed with the pluronic-F-127 gel on the autologous vein grafts can inhibit the proliferation of the vessel proliferating cells,at least partly by inhibiting the adventitia activation,and alleviate the hyperplasia of both neointima and media.
