The effect of ligustrazine on the level of Bax,Bcl-2 and NO,NOS in cerebral ischemia mice after cerebral ischemia reperfusion

Abstract

Objective To explore the effect of ligustrazine on the levels of Bax,Bcl-2 and NO,NOS in cerebral ischemia mice after cerebral ischemia reperfusion(CIR).Methods Totally 72 healthy Kunming mice were randomly divided into sham operation group,ischemia group,ligustrazine intervention group.The biochemical NOS activity and NO content in brain tissue were detected 24h after operation,and the levels of Bax and Bcl-2 protein expression in hippocampus were detected by immunohistochemistry 7d after operation.The histological grade and neuron density(ND) in hippocampal CA1 area were observed with light microscope.Results It showed that the brain NOS activity in ischemic mice(40.7±4.0) U/g pro was higher than those in the sham group(17.0±5.0) U/g pro and the intervention group(31.1±2.4) U/g pro(P0.01);NO content in ischemia group(1.42±0.11)μmol/g pro was higher those in the sham group(0.71±0.14)μmol/g pro and the intervention group(1.03±0.09)μmol/g pro (P0.01).Compared with ischemia group,the Bax expression was significantly reduced in the intervention group (P0.01),while the Bcl-2 expression was significantly increased(P0.01).As for neuron density,the number in sham operation group(144±9/mm) was higher than those in intervention group(96±6/mm) and ischemia group (60±5/mm)(P0.01).Conclusion It proved that the ligustrazine could enhance the ability of brain tissue against oxidative stress,reduce oxygen free radicals injury,thereby inhibiting cell apoptosis and release the reducing the hippocampal neuronal damage induced by cerebral ischemia.
