Abstract

Objective To explore the effects of alcohol on hypoxia-inducible factor and anti-apoptosis protein Survivin in gastric cancer cells. Methods Human gastric cancer cells BGC 823 were separated into 6 groups.5 to 30% final concentration of ethanol were added in,and then the cells were cultured for 1-4 h.A control group as well. After we obtained the cell protein, the HIF-1 α and Survivin protein expression were determined by Western blot analysis. Results In BGC823 cells, the HIF-1 α protein expression increased When the ethanol concentration and the time increasing.Survivin protein expression firstly increased and then decreased with the increase of alcohol concentration when ethanol worked for 1 h.But Survivin protein expression finally reduced with the increase of alcohol concentration when ethanol worked for 4h.Conclusion In BGC823 gastric cancer cells, ethanol induce hypoxia through the HIF-1 α way,and promote apoptosis by inhibiting anti-apoptosis protein survivin.
