Abstract
Objective:To establish the animal models of osteoporosis in ovariectomized rats and investigate the effects of Gujinsan on bone mineral density(BMD) the level of estrogen(E2) in ovariectomized rats.Methods:40 six-month-old Sprague-Dawley female rats were randomly assigned to two groups,including ovariectomized group and false operation group.In the ovariectomized group,double ovaries of rats were removed.In the false operation group,only the fat of rats were exposed.At 1 month after operation,the ovariectomized group were randomly allocated into ovariectomized control group,estrogen group,Gujinsan group,and received medicine intervention as follows: estrogen group: intragastric administrated stilboestrol(0.05mg/kg per day);Gujinsan groups: intragastric administrated Gujinsan(1.8g/kg per day);the rest of groups:intragastric administrated the same amount of normal saline.After 2 months,serum E2 was detected by using enzyme linked immunosorbent assay(ELISA) and BMD was measured by using double functional X-ray digital bone density meter.Results:Compared with false operation group,the body weight of ovariectomized control group was significantly increased(P0.01),and that of ovariectomized control group was also significantly increased compared with that of estrogen group and Gujinsan group(P0.01).BMD and serum E2 of ovariectomized control group were decreased significantly(P0.01),while compared with ovariectomized control group,BMD and E2 of estrogen group and Gujinsan group were increased significantly(P0.05).Conclusion:Gujinsan can increase BMD and serum E2 in ovariectomized rats,which may be one of the mechanisms of Gujinsan in the treatment of osteoporosis.
