Abstract
Objective To investigate the effect of tetramethylpyrazine(TMP)on the proliferation of HSC-T6 cells and the ex-pression of connective tissue growth factor(CTGF).Methods HSC-T6 cells were treated with TMP at different concentrations for various hours,then determined for proliferation by MTT method,for syntheses of collagens types Ⅰand Ⅲ as well as hyaluronic acid(HA)by ELISA,and for expression of CTGF by Western blot.Results The proliferation of HSC-T6 cells was significantly inhibited with TMP at concentrations of 100~1 000 mg /L,and the inhibitory effect was dose-dependent.The syntheses of collagens types Ⅰ,Ⅲ and HA as well as the expression of CTGF decreased.The correlation coefficients(r values)of decreases in syntheses of collagen types Ⅰ,Ⅲ and HA to that in expression of CTGF were 0.727,0.643 and 0.769 respectively,indicating that the decrease in the former was positively correlated to that in the latter.Conclusion TMP inhibited hepatic fibrosis effectively by a potential mechanism of inhibiting the proliferation of HSC-T6 cells and decreasing the expression of CTGF to block the formation of intracellular matrix.
