Abstract
Objective To study the anti-tumor effects of interferon-γ on MGC-803 gastric cancer cell lines and to probe its mechanism.Methods To detect the anti-proliferation and the cytotoxicity of IFN-γ on MGC-803 gastric cancer line by MTT assay.The morphological changes of the apoptosis cell were observed by MTT assay,invert microscope,HE stain, scanning electron microscope.Inducing apoptosis of IFN-γ on MGC-803 gastric cancer cell line was evaluated by TUNEL methods;The positive expression ration of p53、p16、C-myc、Bc1-2 and bax were determined by immunocytochemistry (ICC).Results MTT assay showed that 1250-10000u/ml IFN-γ have the linhibitive action on MGC-803 gastric cancer cell line.MTT stain assay observed that normal cells stain equality, apoptotic cells became small and were stained deeply blue, dead cells became vacuole style and not to be stainde.Morphology showed that cell volume lessened and apoptotic boby sticked to the surface of carcinoma cells in experimental group and apoptotic body formed after effect.TUNEL showed the apoptosis rate increased with time and concerntration.Immunocytochemistry showed that protein expression of p53、p16、C-myc haven't changes obviously(P0.05) and the protein expression of Bcl-2 inclined and Bax increased and have different significantly compared with control respectively(P0.01).Conclusions interferion-γ can inhibite obviously proliferation of MGC-803 gastric cancer cells and induce apoptosis to tumor cells;The mechamism of interferion-γ inducing apoptosis of MGC-803 gastric cancer cells can inhance Bax proteins expression and decline Bcl-2 proteins expression.
