A study on different staining methods to Cyprinus carpio spermatozoon

To select a appropriate method of staining Cyprinus carpio spermatozoon,gentian violet,basic fuchsin,Giemsa,methyl green,hematoxylin eosin Y,eosin Y-ethanol,eosin Y-water,eosin Y-aniline black dye liquors were applied.The results showed that tabletting the spermatozoon with eosin Y before drying was the best method,the staining was clear,sperm was integrity.Staining after natural drying could make Cyprinus carpio spermatozoon′s head membrane rupture cytoplasmic leakage,resulting in sperm staining patterns fuzzy and incomplete.4% formaldehyde fixed before natural drying could avoid carp sperm head membrane rupture.With 1% NaCl diluted semen staining could be clearly observed that the sperm morphology and distinguish abnormal sperm,diluted semen staining could be observed in sperm tail membrane integrity.A supravital eosin Y-aniline dyeing method could be observed head membrane integrity and damage degree of the sperm.
