An immunohistochemical study of prostatic carcinoma with the use of monoclonal antibody against prostate specific membrane antigen

Objective To determine the expression of prostate specific membrane antigen (PSM) in prostate cancer and to investigate the relationship between the PSM expression and histologic grading of prostatic carcinoma and the hormone refractory prostate cancer. Methods 36 cases of prostate cancer, 6 prostatitis, 10 benign prostate hyperplasia (BPH), 10 normal prostate tissue and 38 non prostate neoplasms (including bladder cancer, renal cancer, testis neoplasm, colon rectal cancer,etc.) were analysed for the expression of PSM by using Ed 5 monoclonal antibody and EnVision TM immununohistochemical staining. Results Both benign and neoplastic prostate cells except 3 cases of prostate cancer were stained positively, the staining being intense in hormone refractory lesions.Non prostate neoplasms were stained negatively with the antibody.The relationship between the PSM expression and the histologic grading of prstate carcinoma was difficult to be established. Conclusions The monoclonal antibody Ed 5 was organ specific.It might be used for the imaging and the targeting treatment of prostate cancer.
