Abstract
The HA gene of H5 subtype AIV was amplify by RT-PCR from the DK/Zhejiang/11/00(H5N1) and stepped into pFastBac plasmid,the recombinant plasmid was transforming DH10Bac competent cells to get the positive recombinant bacmid.Recombinant baculovirus were harvest by transfecting bacmid to sf 9 cells.PCR confirmed that HA gene had inserted into the baculovirus genome.Indirect immunofluorescence assay(IFA),SDS-PAGE analysis and Western-blot test showed that H5HA gene has been expressed in sf 9 cells infected with the recombinant baculovirus,and which proved the expressed protein have good biological activity and special immunogenicity.
