Effects of cyclosporine-A immunosupression on allograft nerve regeneration

Abstract

[Objective]To study the effects of immunosupression with cyclosporineA on nerve regeneration in fresh and frozen peripheral nerve allograft.[Method]Thirty-two rats were randomly divided into two groups.In each animal,bilateral 10mm sciatic nerve defects was made,the left sciatic nerve was bridged with fresh nerve allograft and the right side with nerve allograft frozen in liquid nitrogen.The rats in group B were given cyclosporine A.Eight weeks after operation;electrophysiological and histological assay were conducted in each rat.[Result]Nerve regeneration in fresh allograft without immunosupression was the worst among these groups;in contrast,it was the best in fresh allograft under immunosupression with CSA.Nerve regeneration in frozen nerve allograft with CSA was better than that without immunosupression.[Conclusion]Immunosupression with cyclosporine-A is helpful to suppress the rejection of nerve allograft and to improve the peripheral nerve regeneration.
