Abstract

Objective To study the expression and the relationship between NF-κB and TNF-α by rats pulmonary alveolar macrophage(PAM) stimulated by Aspergillus fumigatus(A.fumigatus) and the role of TLR4 in it.Methods PAM from Wistar rats were served in vitro as target cells treated with different dose of A.fumigatus(5× 104, 1×105 and 2×105) and anti-TLR4 mAb(5 μg), single or both to measure the levels of NF-κB and TNF-α.Results The levels of TNF-α were significantly increased after stimulated by A.fumigatus conidia for 0.5 h,1 h,2 h and dose-dependently higher than that of in the negative team(P 0.05), anti-TLR4 could decrease them partly;the lev-els of NF-κB were increased after stimulated by A.fumigatus conidia for 0.5 h, reached the highest level in 1 h later and then decreaed, anti-TLR4 could decrease them partly.Conclusion NF-κB signal transducting pathway mediated by TLR4 may play an important role in the production of TNF-α by PAM stimulated by A.fumigatus conidia.
