Abstract
Objective: Severe acute pancreatitis(SAP) models of rats were set up to investigate the exspression of CXCL11 in the lung.The effects of triptolide were evaluated by inhabiting the expression of CXCL11 in AP.Methods:Seventy-Two SD rats were randomly divided into three groups: the control group(C group),the AP model group(P group),and the Triptolide treatment group(T group).and there were 24 rats in each group.SAP group was established by 1ml/kg retrograde infusion of 4% sodium taurocholate into the bili-pancreas duct in SD rats.N group was not injected with 4% sodium taurocholate.In T group,triptolide was injected into the abdominal cavity immediately after the SAP model was achieved.each group was randomly divided into four sub-groups and each sub-group contained 6 rats.The rats were randomly selected to be executed at 1,3,6 and 12 hours later in each group.Serum was amylased,and the wet/dry ratio of the lung and the pathological changes of the lung were measured at different time-point in each group.Protein in the lung was decected by immunohistochemistry.Expression of CXCL11 protein in serum was measured by enzyme-linked immunoadsorbent assay.Results:(1)Serum amylase:serum amylase was significantly higher in P group and Tgroup than in N group at each time point(P0.01).And amylase of 6h and 12h subgroups in T group was lower than in P group.(2)Lung wet/dry weight ratios:they were significantly higher at 3h,6h and 12h in P group and at 6h and 12h in T group compared with N group.(3)Immunohistrochemistry: Damages of lung tissue at 3h,6h and 12h of P group and T group were significantly higher while at 6h and 12h the damage of T group was slighter compared with P group.T group was slightly lower than P group in expression of CXCL11.(4)ELISA:serum CXCL11 at 1h,3h and 6h in P group was higher than in N group and T group and at 12h no significant difference ocurred when P group was compared with N group and T group.Conclusions:Triptolide may ameliorate acute lung injury by inhibiting the activity of CXCL11 and anti-inflammatory action in SAP.
