Abstract
Cold Fraction IV(FⅣ) paste contains 96.00±13.52g /kg of albumin. 87.44 percent of the albumin (about 83.94±11.11g/kg) could be dissolved in 15 fold volumes of PBS saline from FⅣ pasre, of which 48.18±1.11g/kg of albumin could be recovered by a two step process with a DEAE Affi Gel Blue affinity chromatography and a DEAE Sepharose FF ion exchange chromatography. The recovery rate is 57.40±1.94%(to Fiv solution). The albumin preparation shows a little yellow colour, and is clear. The purity of albumin is 98.28±1.68%, monomer is 97.00±2.1%, and Abs1% 403nm icm 0.028±0.006. The albumin preparation is stable after heat treat at 57℃ for 50 hours. All biochemical index of albumin preparation purified from Fiv paste is capable of meeting the requirement in “Chinese Regulation of biological Products”.
