Abstract

Try to find the effect of lead on memory and the expression of calsyntenin-2 mRNA in hippocampus of mice,and provide scientific evidence for further revealing nerve poison mechanism of lead.Pregnant mice were divided into three groups of 10 each,were exposed to 0.3、1.0 and 3.0 g/L Pb acetate in drinking water from the first day of gestation to postnatal day(PND)21,to determine the lead concentration in blood and hippocampus by graphite furnace atomic absorption spectrometry,the ability of learning and memory and the expression of calsyntenin-2 mRNA in hippocampus by Y-type maze and RT-PCR at 21 d post treatment.The lead concentration in lead exposure group was higher than control group(P0.05).The result of Y-type maze demonstrated,the ability of learning(T1),memory(T2) and the memory retaining rate(T3) in 21-day old offspring in the three lead exposed groups were significantly decreased(P0.05).The relative expression level of calsyntenin-2 mRNA in 0.3 g/L Pb exposure group had no significant diference with control group(F=13.216,P0.05).However,there was obviously statistical significance between 1.0 g/L and 3.0 g/L lead groups(F=53.247,P0.05;F=96.318,P0.05).Maternal lead exposure may induce the accumulation of lead,the decreased expression of calsyntenin-2 mRNA in hippocampus of the offspring,which resulted in learning and memory damages.
