Abstract

Objective: Construction of TRAIL expression of engineering strain in order to obtain biologically active recombinant protein.Method:TRAIL gene was expressed in E.coli successfully after IPTG induced.Then the soluble forms of protein was increased by Optimization of the host and the temperature.TRAIL was purified by ammonium sulphate precipitation,Ni Sepharose 4B,Q sepharose FF and Source 30 S and analyzed its activity in vitro.Result: TRAIL was up to 30% of the total bacterial protein after IPTG induction,furtherlly inctrased to 55% of the soluble form of protein after optimization.The purified TRAIL can inhibit a variety of tumor cells proliferation,with inhibition rate of 70%-92% in vitro tests.Conclusion: Recombinant TRAIL was highly expressed in soluble form,and has good biological activity,with the protein for its development lay the foundation for medicine.
